Mouse models for the in vivo validation of protein interactions

In subproject 8 mouse models are generated which express distinct proteins tagged with a fluorescence dye and therefore can be easily detected and isolated. Here, we work on disease related proteins and the tagged protein is isolated from several tissues of mice. By using mass spectrometry of the protein complexes the identity of the proteins can be determined and the network of proteins, which contribute to a biological function or the evolving of a disease specific to the organ can be revealed. Subproject 8 takes a central position within the DiGtoP consortium as it validates the interaction profiles of the proteins in vivo for the first time and in addition creates mouse models for the DiGtoP consortium, collaboration partner, and NGFNplus members. Due to the results generated up to now, the principle of the proteomic analysis could be confirmed in vivo. Thus, interaction partners already known from cell cultures have been confirmed, but also new interaction partners as well as organ specific differences have been detected.
